Objective:
1. Improve exosome purification via size exclusion chromatograph/RNase/protease treatments. 2. Catalog/characterize exosomal sRNAs and proteins from exosomes from control and treated (FHB/trichothecene) barley seedlings. 3. Use super resolution microscopy to image exosomes and measure impact of isolated exosomes on fungal growth (in vitro).

Approach:
We have successfully isolated barley exosomes using ultracentrifugation and density gradients. Our previous proteomic analysis indicated that additional purification (size exclusion chromatography and/or RNase/proteinase treatment) is necessary to improve the quality of our proteomic and genomics results. We have all the tools needed to meet the three objectives listed above.
