Interpretive Summary: Soybean cyst nematodes(SCN) are a major pest of soybean in the USA and world-wide. Soybean breeders have developed cultivars with genetic resistance to several of the races of SCN. In this paper we characterized specific regions of soybean chromosomes for SCN resistance and evaluated several molecular markers previously associated with SCN resistance. One marker in particular, pBLT65 was of significant value for tracking SCN resistance on soybean chromosome linkage group A. This information is important to soybean breeders attempting to move SCN resistance from one soybean cultivar to another.

Technical Abstract: One of the most destructive pests of soybean, Glycine max, in the United States is the soybean cyst nematode (SCN), Heterodera glycines. In order to identify molecular markers with significant linkage to chromosomal regions containing resistance to SCN, one hundred twenty-five F3 progeny were evaluated from a cross between J87-233 and SCN susceptible 'Hutcheson' Soybeans. Plants were evaluated for resistance to SCN races 1,2,3,5, and 14. RFLP analysis used 215 genomic clones. RAPD analysis included 45 primer combinations. Linkage between pBLT65 and the QTL on linkage group A of the soybean linkage map was significant for SCN.
