Publication Type: Abstract Only Publication Acceptance Date: 10/1/1996 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: Chicken cDNAs encoding interferon-gamma (IFN-g) and IL-15 were cloned from concanavalin A (Con A) activated spleen lymphocytes using reverse transcription-polymerase chain reaction (RT-PCR) with oligonucleotide primers constructed based on published sequences. The expression of IFN-g and IL-15 was determined by Northern hybridization and RT-PCR. The sequences of IFN-g and IL-15 were verified by dideoxy chain termination and automatic sequencer. The message of IFN-g was not detected in heart, liver, spleen and oviduct. The expression of IFN-g was detected as early as 30 min after Con A activation of spleen lymphocytes and reached a peak expression at 2 hr activation, persisting to 4 hr followed by sharp decline thereafter, indicating that IFN-g may be an early expressed gene upon mitogenic stimulation. The message for IL-15, unlike IFN-g, was detected in various tissues, including heart, liver, spleen, oviduct and Con A activated spleen lymphocytes. It is in agreement with the report that described the human IL-15 is expressed in various tissues. These results will make it possible to understand the function of cytokines on the chicken immune system. |
