Interpretive Summary:

Technical Abstract: The chicken MHC haplotype (B complex) is generally determined by the use of alloantisera in hemagglutination assays (HA). The haplotypes B12 and B13 are highly cross-reactive. This cross-reactivity could be due to shared antigenic epitopes in either B-G, or B-F (class I) antigens. DNA from the BFIV locus of the B12 and B13 haplotypes was cloned and sequenced. The predicted amino acid sequences of these two alleles were compared with amino acid sequences available from five other BFIV alleles. Amino acid polymorphisms that occurred only in B12 and B13 were identified. These amino acids (147-155) are predicted to be within the alpha helix of the class I molecule and exposed for antibody elicitation and interaction. BFIV12 and 13 alleles were inserted into the RCAS retroviral expression vector and expressed in an avian B-cell line (RP9). Alloantisera analysis by flow cytometry of the cell lines expressing the BFIV12 or 13 alleles showed the same cross-reactivity patterns as seen by HA suggesting that the class I alleles from BFIV are important in this cross-reactivity. In vitro mutagenesis studies in which amino acids 147-155 were altered confirmed the significance of this region for B12 and B13 alloantiserum cross-reactivities.
