Interpretive Summary: This manuscript summarizes the results of part of the second round of analyses performed by scientists participating in the First International Swine CD Workshop. This workshop was set up to compare and standardize the reactivities of monoclonal antibodies (mAb) that recognize pig leukocyte cell surface markers to assign them specific cluster of differentiation numbers (CD#). Investigators from 8 laboratories analyzed the reactivity of a panel of 26 monoclonal antibodies (mAb) that had been identified by earlier analyses possibly to be reactive against porcine CD44 or CD45. An anti-human CD44 mAb (Z062) was added because it exhibited positive cross reactivity with porcine CD44. Final results from testing of this CD44/CD45 workshop panel established the assignment of 7 mAb as reactive with CD44. Several CD44 clustered mAbs were examined in more detail to determine whether they bound to the same area, or epitope, of the CD antigen. Two of these mAb appear to react with the same epitope, however, immunoprecipitation to define the antigen mw has not yet been conducted and so they are designated as reactive with wCD44a. Four mAb were shown to be reactive with CD45 and 4 other mAb as reactive with the more restricted CD45R epitope. These CD45 assignments were based on flow cytometric staining parameters and on molecular weights of the porcine antigens precipitated by the mAb. Many mAb from the CD44/CD45 panel went unassigned, these mAb will be assigned to the non-lineage specific panel for the Second CD Workshop and will require more detailed molecular weight and functional analyses. This panel of 7 CD44 and 8 CD45/CD45R specific mAb will enable porcine immunologists to better characterize subsets of pig lymphoid cells.
