Technical Abstract: Using a modification of a previously developed sandwich ELISA procedure, we determined the amount of decorin core-protein (DCP) content of raw powdered hide and powdered hide treated with the early steps of the tanning process. We found approximately 0.15 mg DCP/g hide in raw powdered hide. Treatment with reliming reagents results in a drop of approximately 90% in available DCP content. Deliming and bating had no measurable effect in further removing DCP from the powdered hide. The results with hide powder suggest that the early steps of hide processing can remove decorin. Nevertheless, diffusion rates may limit such removal in intact hides, thereby suggesting the next round of experiments in this project aiming to correlate decorin removal with leather properties.
