Interpretive Summary:

Technical Abstract: Breeding for resistance to Beet curly top virus (BCTV) has been difficult because screening of selected lines in the field results in usually sporadic non-uniform infections across and within test plots. In order to circumvent the lengthy process required for identification of genetic resistance in bean, DNA markers have been developed that are tightly linked to the BCTV resistance gene(s) and have applications for marker-assisted selection (MAS). A RAPD marker directly linked (0.0 cM) with a dominantly inherited BCTV resistance gene was identified in a recombinant inbred snap bean population (Moncayo/Primo) consisting of 94 F5:7 lines that had uniform segregation for disease reaction to BCTV across three field locations. The RAPD marker was converted to a SCAR marker and mapped within a cluster of disease resistance genes on linkage group B7 of the core map. A survey of 112 BCTV-resistant and susceptible bean cultivars and breeding lines revealed the SCAR would be highly useful for MAS of the resistance gene in snap bean and dry bean of Andean origin.
