Interpretive Summary:

Technical Abstract: Interleukin-16 is a proinflammatory cytokine which is synthesized as a precursor protein (pro-IL-16). Two EST clones (G24, J10) which are homologous to mammalian IL-16s were identified from chicken intestinal cDNA library which we have established recently. Chicken Il-16 gene contains the entire open reading frame (ORF) of pro-IL-16 and encodes the mature IL-16 protein which consists of 607 amino acids. Chicken IL-16 shows 86% sequence homology to duck pro-IL-16 and 49-52% to various mammalian IL-16s. By Northern blot analysis, IL-16 transcripts showed a restricted expression to the lymphoid tissues. Recombinant chicken IL-16 which is consisted of 149 C-terminal amino acids of pro-IL-16 is biologically active when expressed in COS-7 cells and showed chemoattractant property for lymphocytes.
