Interpretive Summary:

Technical Abstract: Monitoring of patulin in apple juice, apple juice concentrates and apple juice products is performed to comply with guidelines of the United States Food and Drug Administration. The majority of the current methods are based on AOAC Official Methods involving liquid-liquid extraction of patulin with ethyl acetate, followed by high-performance liquid chromatography for detection and quantification. Samples of apple juice and apple juice concentrates were purchased for analysis. Four different extraction methods were compared: ethyl acetate liquid-liquid extraction (AOAC Official Method 995.10) and three solid-phase extraction cartridges (Oasis HLB, Strata X and MycoSep 228). Analysis was by HPLC with photodiode array detection. To obtain separation of patulin and 5-hydroxymethylfurfural, with minimal presence of co-extracted compounds, different mobile phases and flow rates were compared. Use of solid phase cartridges required fewer steps, but careful control of column flow was required. Results of the different methods were similar in both the amount of patulin detected and the recoveries in spiked samples. None of the samples analyzed contained patulin in excess of the FDA regulated level for patulin of 50 ug/l.
