Technical Abstract: Previous attempts at separating nonpolar lipid esters (including wax esters, and methyl esters)have only achieved limited success (Moreau & Christie, 1999). We recently found that wax esters co-chromatograph with phytosteryl esters in our previously reported normal-phase HPLC method (Moreau et al., 1996). Several normal phase methods have attempted to improve this separation (Inger-Elfman-Borjesson & Harrod, 1997; El-Hamdy & Christie, 1993; Foglia & Jones, 1997). Recently a report of a method employing an alumina column at 30 degrees C, described promising results at separating these nonpolar lipid classes (Nordback & Lundberg, 1999). In the current study, modification of the alumina method by increasing the column temperature to 75 degree C improved the separation of standards of wax esters and sterol esters.
