Interpretive Summary:

Technical Abstract: Commercial bacterins for the control of fowl cholera are comprised of P. multocida serotypes A:1, A:3, and A:4, which are the serotypes that are most often associated with outbreaks in poultry flocks. The 39 kDa antigen was previously isolated from in vivo-grown P. multocida and was found to induce active immune cross protection and passive immune cross protection depending on the dose administered. An outer membrane protein extract containing the 39 kDa was submitted to denaturing 1-dimensional sodium dodecyl electro-phoresis. The 39 kDa protein was identified from a plot of molecular weight versus mobility and excised from the Coomassie Brilliant Blue-stained gel. The excised band was digested with trypsin, and the peptides purified by reverse-phase HPLC. Selected peptide peaks were analyzed by N-terminal sequencing, MALDI-TOF mass spectrometry, and database analysis using MS-FIT. The MS-FIT data were confirmed by N-terminal sequence information for two peptides of the digest. The results indicated that the 39 kDa cross protective antigen is the P. multocida lipoprotein B, PlpB.
