Interpretive Summary: Technical Abstract: The gene sequence for a 47 kDa surface protein of Cryptosporidium parvum sporozoites that has homology to ankyrins was expressed as a recombinant polyHis fusion protein. As measured by indirect immunofluorescence (IFA) staining, antisera against rCP47 bound a surface antigen of C. parvum sporozoites and cross-reacted with an antigen on the oocyst wall. Methanol ltreatment of sporozoites prior to IFA staining enhanced the immunofluorescence signal suggesting that native CP47 is an integral membrane protein. Cell cultures infected with excysted sporozoites that had been pre-treated with anti-rCP47 sera showed lower parasite development compared to treatment with pre-bleed serum as measured by semi-quantitative PCR. Studies are underway to produce hyperimmune bovine colostrum (HBC) against rCP47 for passive immunotherapy against cryptosporidiosis in newborn calves. |
