1) Compare differing plant-based assays for cost efficiency and their ability to detect resistance to transgenic toxins targeting rootworms. 2) Assist in evaluating current artificial diets for western corn rootworm growth and development.
For Objective 1, we currently have three laboratory colonies selected for resistance to Cry3Bb1, one to mCry3A, at least one to Cry34/35Ab1, and one to eCry3.Ab. Each of these colonies has at least one paired unselected control colony. In addition, we have three field populations with resistance to Cry3Bb1 and one field population susceptible to all transgenic toxins targeting WCR. Finally, we have also used colonies from the USDA-ARS lab in Brookings, SD as susceptible controls. Each selected and control colony will be evaluated on both the Bt to which the selected colony was selected on and its corresponding isoline in each of the reported plant-based assays for evaluating WCR transgenic proteins. The efficiency of each assay to detect resistance will be compared and then the labor required for each will be calculated. Although, standard techniques will be utilized, the experiment will be time consuming and somewhat labor intensive. For Objective 2, we will evaluate survivorship and growth of WCR larvae on artificial diets to determine which diet provides the best rearing conditions for WCR. Published techniques will be utilized under antiseptic conditions.
