Technical Abstract: Hypoxanthine guanine phosphoribosyltransferase (HPRT1) has been mapped to human chromosome Xq26-q27 and assigned to porcine chromosome X with somatic cell hybrids. We have mapped HPRT1 to a linkage group on porcine X chromosome using an RFLP and linkage analysis. A total of four RFLPs was identified with the HPRT1 cDNA probe. Polymorphisms identified by EcoRI and BglII followed X-linked inheritance and mapped to one locus on chromosome X. Polymorphisms defined by BamHI and PstI were autosomally inherited, mapped to chromosome 7 by two point linkage analysis and must represent either HPRT2 or one of 4 pseudogenes present in man. Position of both loci within a linkage group was determined by multipoint linkage analysis. No recombination was detected between HPRT1 and the microsatellite SW2453 which has been physically assigned to Xq25 by in situ hybridization. Comparative mapping provides gene assignments to specific chromosomes and additional information to define the gene order. Assignment of HPRT1 gene in swine provides a reference locus for alignment of X chromosomes between porcine and other species such as human. Our results concur with the results of synteny mapping between humans and pigs by the methods of heterologous chromosomal painting where human chromosome X is found to be homologous to pig chromosome X.
