Inoculation and growth with foliar pathogenic fungi. In: The Medicago truncatula Handbook. The Samuel Roberts Noble Foundation, Ardmore, OK. Available: http://www.noble.org/MedicagoHandbook/. Interpretive Summary: Technical Abstract: This chapter describes methods for evaluating responses of Medicago truncatula to foliar plant-pathogenic fungi. Growth of plants, preparation of inoculum, incubation conditions, and disease assessment are described for five necrotrophic pathogens (Ascochyta lentis, Ascochyta rabiei, Colletotrichum trifolii, Mycosphaerella pinodes, Phoma medicaginis) and one biotroph (Eysiphe pisi). For several diseases both whole plant and detached leaf methods have been developed. For each assay, check plants and their responses to the pathogen are listed. Methods for re-isolation of the pathogen from diseased tissue and storage of cultures are also detailed. For several diseases, methods for staining and visualization of the fungi in plant material are described. |
