Interpretive Summary: Charcoal rot, a disease of global importance, has become a disease of major impact throughout the soybean production regions of the USA with an estimated yield loss of 10-50% in infected areas. This disease, greatly favored by hot, dry soil conditions, is essentially impossible for growers to manage due to the lack of pesticide treatments to combat the fungus, and the lack of resistant cultivars. Scientists at the USDA-ARS located in Stoneville, MS have identified and released a soybean line with moderate resistance to charcoal rot. This provides, for the first time, genetic resistance to charcoal rot available to soybean breeders in the public and private sectors serving the 75 million acres produced in the USA.

Technical Abstract: ‘DT97-4290’ soybean [Glycine max (L.) Merr.] was developed as a maturity group IV germplasm line by the USDA-ARS, Stoneville, MS. It was released in December 2004 because of its high yield potential and moderate resistance to charcoal rot [caused by Macrophomina phaseolina (Tassi) Goid.] and southern stem canker [caused by Diaporthe phaseolorum (Cooke & Ellis) Sacc. var. meridionalis F.A. Fernandez]. DT97-4290 originated as an F5 single plant selection from the cross: Asgrow ‘A5979’ × Delta Pine ‘DP3478’. DT97-4290 has an indeterminate growth habit, purple flowers, tawny pubescence, and tan pod walls. Seed are shiny yellow with black hila. DT97-4290 is classified as Maturity Group IV (relative maturity 4.8), and matures about 2 d earlier than ‘Manokin’. In three years of USDA Uniform tests, plant height of DT97-4290 averaged 91 cm compared to 75 cm for that of Manokin. The plant lodging score of 2.0 (where 1 = all plants upright and 5 = all plants prostrate) and seed quality score of 2.2 (where 1 = excellent and 5 = poor) of DT97-4290 were similar to those of Manokin. Seed weight of DT97-4290 averaged 144 mg/seed compared to 122 mg/seed for Manokin. Seed of DT97-4290 averaged 415 g/kg protein and 197 g/kg oil which is 11 g/kg more protein and 9 g/kg less oil than that of Manokin. In 38 southern environments, seed yield of DT97-4290 averaged 3176 kg/ha which is 2 % greater than that of Manokin. In 15 environments, five locations during three years, in the Mississippi State University soybean variety trials, DT97-4290 averaged 3169 kg/ha which was 97% of the test mean. DT97-4290 is moderately resistant to charcoal rot based on field evaluations from 2002 to 2004 in artificially infested sandy loam soil at Stoneville, MS. Host tissue colonization by M. phaseolina was assessed in three replications by destructively sampling ten plants at the R7 growth stage. The severity of internal discoloration for stem and root was measured on a scale of 1-5 (where 1= resistant and 5= susceptible). The disease severity ratings over the three years averaged 1.6 and 3.6 for DT97-4290 and Manokin, respectively, with 4,388 and 8,331 colony forming units for DT97-4290 and Manokin, respectively. DT97-4290 is also moderately resistant to frogeye leaf spot. DT97-4290 is resistant to southern stem canker, soybean mosaic virus, and races 2, 4, and 10 of phytophthora rot [caused by Phytophthora sojae (Kaufmann & Gerdemann)].
