Technical Abstract: cDNA clones (1.9 kb in length) have been developed for two HSP70 transcripts (HSP70A and HSP70B) using RT-PCR and 3'-RACE. The clones share 70% identity at the nucleic acid level. Both HSP70A and B are induced by exposure to 39 deg. C for 1 hour. HSP70A, but not HSP70B, is expressed as a normal part of the diapause program. HSP70A transcript is detectable within 3 days of adult burrowing and persists for at least 4 months at normal rearing temperatures. Exposing diapausing beetles to a thermoperiod with a mean temperature of -2.5 deg. C results in a high level of expression of HSP70B, but only a minor increase in the expression of HSP70A. This study has clearly demonstrated that members of the 70 kDa heat shock protein family are differentially expressed during normal overwintering conditions in the Colorado potato beetle.
