Technical Abstract: A North Dakota strain of the Colorado potato beetle (CPB), Leptinotarsa decemlineata (Say), was reared under short - (8L:16D at 20 C) and long-day (16L:8D at 25 C) conditions. Hemolymph proteins and free amino acids were examined in both sexes over time and after pyriproxyfen (JHA) treatment in CPB held under both long- and short-day conditions. Pre-diapausing and diapausing adults contained 2-to-3 times more protein than non-diapausing adults did. Total free amino acid concentration in the hemolymph increased gradually up to 20 days of the adult life under short-days, but increased markedly during the first 10 days of the adult stage and then decreased under long-day conditions. Proline and glutamine were the most abundant free amino acids in CPB held under either long- or short-day conditions.. JHA treatment to diapausing adults held under both long- and short-day conditions terminated diapause as indicated by re-emergence from the vermiculite, feeding, and mating with the reduction of total protein concentrations, elimination of diapause protein 1 and the appearance of vitellogenin. In addition, CPB held under long days also matured oocytes and oviposited. Short-day beetles showed an increase in total free amino acid concentration of 20% in females and 38% in males after JHA treatment. However, both sexes held under long-days after treatment showed a slight decrease in total amino acid levels. Proline and glutamine, the major free amino acids, increased after JHA treatment under short-day conditions but, decreased under long-day conditions.
