Interpretive Summary: Red rot, caused by the fungus Colletotrichum falcatum, is a destructive disease prevalent in most sugarcane-producing countries. Disease-free sugarcane planting materials are essential as the pathogen spreads primarily through infected setts. The present study reports the development of a diagnostic technique known as loop-mediated isothermal amplification or LAMP to detect the pathogenic fungus in sugarcane setts used to plant a new crop of sugarcane. The LAMP assay is convenient, rapid, and economical because results are visual and easy to interpret, the test can be conducted in less than an hour, the assay is more sensitive than most others, and specialized equipment is not required. This is the first report of LAMP-based detection of the red rot causing pathogen in sugarcane setts. Technical Abstract: Red rot, caused by Colletotrichum falcatum, is a destructive disease prevalent in most sugarcane-producing countries. Disease-free sugarcane planting materials are essential as the pathogen spreads primarily through infected setts. The present study was undertaken to develop loop-mediated isothermal amplification (LAMP) detection of C. falcatum genomic DNA with four sets of specific primers. Under isothermal conditions, DNA is amplified with high specificity, efficiency, and rapidity. Moreover, visual judgment of color change in <1 hr and without the need for a thermocycler makes the method convenient and economical. With C. falcatum DNA, sensitivity of the LAMP method was observed ten times higher than that of conventional PCR. This is the first report of LAMP-based detection of the red rot causing pathogen in sugarcane setts. |
