Objective:
To determine fine mapping and functional characterization of bacterial wilt disease resistance gene (Bw) in cucumber.

Approach:
1. ARS will use map-based cloning strategy to clone and characterize Bw resistance gene in the cucumber line WI2757. Near isogenic lines (NILs) of Bw will be developed through marker-assisted backcrossing and phenotyping. Recombinants will be identified through flanking markers until the target gene region is narrowed down to less than 50kb. 2. Candidate gene in target region will be identified and annotated. 3. Transcriptome profiling will be performed in NILs before and after pathogen inoculation.
