Objective:
1. Develop and characterize cucumber core population (390 cucumber lines). 2. Molecular mapping of downy mildew (DM) resistances in cucumber inbred line PT108. 3. Marker-assisted pyramiding of DM resistance QTL and evaluation of enhanced germplasm with multiple disease resistances.

Approach:
Objective 1: 1) Re-sequencing and analyze the data for 384 cucumber lines. 2) Phenotype for target traits in this collection for downy mildew resistance. 3) Conduct association analysis for downy mildew resistance. Objective 2: 1) Fine mapping dm4.1, dm5.2, and dm5.3 major-effect QTL. 2) QTL mapping of DM resistance in PT108 from exotic source. Objective 3: 3). Marker-assisted QTL pyramiding of dm4.1, dm5.2 and dm5.3 into elite pickling cucumber background Gy14. Evaluate its performance in field and growth chambers.
