Technical Abstract: Effective disease management in plants depends on timely and accurate pathogen identification. Most plant virus detection methods are based on virus-specific serological (i.e., ELISA) or molecular properties (PCR or real-time PCR). I will demonstrate the development and application of immunocapture Real-time RT-PCR systems for tomato virus survey. I will also discuss application of deep sequencing and assembly of small RNA technology for virus (pepino mosaic virus) and viroid (potato spindle tuber viroid) identification in tomato. Using this technology, we were able to identify a novel potyvirus without prior knowledge and then obtain its complete genome sequence for the first time.
