Interpretive Summary:

Technical Abstract: Expressed sequence tags (ESTs) from 47 wheat cDNA libraries have been mapped to wheat 'Chinese Spring' chromosome group 4. A total of 1670 ESTs with 2259 corresponding EST loci were mapped to all three genomes. The distribution of the mapped ESTs among the three homoeologous chromosomes was not the same, with 4B having the fewest EST loci mapped (643) compared to 4A (880) and 4D (713). This was opposite to what was actually observed when the total number of EST loci in the B genome (5857) was compared to those present in the A (5216) and D (5294) genomes. One possible explanation of this phenomenon could be that 4B contains a higher level of heterochromatin than 4A and 4D. The long arms contained more ESTs than the short arms and the proximal regions showed a higher number of ESTs than the distal regions. This study confirmed the complex structure of wheat chromosome 4A with the occurrence of two reciprocal translocations involving chromosome arm 4AL, 5AL, and 7BS as previsouly identified, but also indicated that two additional inversions occurred in the distal part of 4AS and the centromeric region of 4A. A consensus map among the genomes was developed based on the 321 unique ESTs for chromosome group 4 that only mapped to group 4 chromosomes. Good co-linearity was shown between wheat ESTs on homoeologous group 4 chromosome and rice chromosome 3. A few ESTs also showed homology with rice chromosomes 11 and 12.
