Interpretive Summary:

Technical Abstract: Cytoplasmic organelle specific genes have been associated with many economically important traits including male sterile genes, fertility restorer genes and also genes responsible for susceptibility to insects, diseases and environmental stresses in cotton. However, there is no report at the molecular level, as per our knowledge, on any cytoplasmic-specific genes in cotton. The objective of this research was to identify cytoplasmic-specific transcripts using bulked samples of hybrids from reciprocal crosses of the maternally-inherited morphological mutant (cyt-V). This mutant was used in reciprocal crosses with two different lines Pima S-6 and the virescent mutant (v7v7), respectively. The cDNAs were constructed, using a commercial kit, from bulked samples of the third yellow- colored leaves from the reciprocally crossed plants. An automated PCR-based AFLP method was used to screen the differentially expressed genes in the cDNAs. Six cyt-V specific transcripts, ranging from 100 bp to 500 bp, were identified indicating the possibility that this mutant influences more than one gene in plant development. Several of these transcripts were isolated and the partial sequence of these cytoplasmic-specific cDNAs and their homology with other genes based on BLAST searches will be presented.
