Research Project: Assemble the Alfalfa RegenSY27x Reference Genome

Location: Plant Science Research

Project Number: 5062-21500-001-002-S
Project Type: Non-Assistance Cooperative Agreement

Start Date: Sep 1, 2019
End Date: Aug 31, 2024

Objective:
The objective of the research is to assemble a high-quality, well-annotated de novo reference genome sequence for alfalfa.

Approach:
High molecular weight DNA will be isolated from an alfalfa plant, which is used in many labs around the world for genetic transformation studies. The DNA will be prepared for sequencing using commercially available technology to generate long continuous DNA sequences. Sequencing will be done to 30x coverage. Assembly of the sequences into larger scaffolds will be done using a combination of technologies such as 10X Genomics assemblies, genome mapping, and other commercially available services. Genetic maps may also be used to assemble sequences into longer scaffolds. Annotation of genes will be done using appropriate software and transcriptome data.
