Technical Abstract: Paratuberculosis (Johne's disease) caused by Mycobacterium avium subsp. paratuberculosis (MAP) is a serious economic threat worldwide affecting cattle, sheep and goats. The infection can persist for several years in a subclinical phase that is difficult to identify because of the absence of clinical signs. Diagnosis of the disease is also complicated by this slow progression of the disease and the prevalence of genetically similar organisms in the environment. Several MAP protein candidates have been evaluated as diagnostic reagents. Yet, large scale proteome analysis from multiple isolates grown under stress conditions are lacking.Though the strains affecting cattle and sheep share some common antigens, their genome contents and growth characteristics vary significantly. In the present study, we hypothesized that the proteomic profiles of cattle and sheep strains of MAP vary with their phenotypic changes when they are grown in-vitro under high iron, low iron and oxidative stress conditions. We studied a cow and a sheep strain of MAP, cultured under nutrient rich conditions (Middlebrook 7H9 medium with Mycobactin J), acute iron depletion (Middlebrook 7H9 medium devoid of iron and Mycobactin J) and in the presence of hydrogen peroxide (10 mM). The relative protein expression levels between the samples were analyzed by iTRAQ using amine-labelled isobaric tags. The data is being analyzed for this presentation.
