Technical Abstract: Yellow dwarf viruses (BYDV and CYDV), spread by aphid vectors, are among the most serious diseases of cereals worldwide. Wheat-Thinopyrum intermedium T7DS.7DL-7EL translocation lines P961341 and P98134 carry Bdv3 for resistance to YDV that originates on the long arm of chromosome 7E. To facilitate efficient screening of wheat populations segregating for this YDV resistance, a diagnostic molecular marker, SSR-Bdv3, which is proximally linked to Bdv3 has been produced in this study. Parent-progeny transmission of the Th. intermedium segment containing Bdv3 was investigated using the SSR-Bdv3 marker and testing for virus titer using ELISA. The transmission frequency was determined in F2 populations and testcross F1 populations derived by crossing each of the above two wheat-Th. intermedium translocation lines to four bread wheat cultivars. The Th. intermedium chromosome 7E segment was preferentially transmitted through male gametes of singlecross F1 plants in all of the crosses and their reciprocal crosses with the four wheat cultivars. Transmission frequency of the Th. intermedium chromosome segment in P961341 varied in different genetic backgrounds. The F2 populations from the singlecross, CS/P961341, and its reciprocal cross showed a good fit to the expected ratio 1 : 2 : 1.In this study, preferential transmission in male gametes for either the 7E segment or the normal chromosome 7D was observed in progenies derived by crossing P961341 to different bread wheat cultivars.
