Interpretive Summary:

Technical Abstract: A general procedure for two-dimensional (2-D) gel electrophoresis of Citrus leaves has been developed. We evaluated the resolution of 2-D protein patterns for Tris-HCl and KCl extractions of water soluble proteins and a phenol extraction for extraction of hydrophobic cell membrane proteins from Chandler pummelo (Citrus grandis) leaves. All three extractions provided satisfactory results. In addition, the versatility of the Tris-HCl extraction method was demonstrated by extracting leaves from six different members of the Citrus family (C, aurantium, Bouquet des fleurs (sour orange); C. tangerina cv. Dancy (mandarin); C. medica, cv. Diamante (citron); C. limon, cv. Frost eureka (lemon); C. sinensis, cv. Powell (navel orange); and C. paradisi, Redblush (grapefruit)).
