Interpretive Summary:

Technical Abstract: BVDV consists of two recognized biotypes, noncytopathic and cytopathic, based on the formation of cytopathic effect in susceptible cells in culture. In BVDV2, the major difference between the two biotypes is the presence in cytopathic strains of an in-frame recombination event resulting in the insertion of a cellular RNA into the NS2/3 region of the genomic RNA. Expression of the inserted sequences leads to the proteolytic cleavage of the NS2/3 to the NS2 and NS3 proteins. PCR amplification using primers designed from the sequence of the genomic insert in BVDV strain 125c yielded multiple bands from uninfected cellular RNA isolated from a number of bovine tissues. Sequences derived from these bands showed potential altered processing of the transcript in this region. Northern blot analysis showed differences in expression of this gene between infected and uninfected cells. Cloning and sequencing of a cDNA clone using the insert sequences as probe revealed that this transcript encodes a dnaJ-like protein, the regulatory protein of a molecular chaperone complex which may also be involved in transmembrane transport.
