Objective:
Develop barley/cereal yellow dwarf virus resistant barley germplasm.
1) Examine the prevalence of strain composition of barley/cereal yellow dwarf viruses on barley and wheat in Nebraska.
2) Identify the aphid fauna in barley and wheat fields in Nebraska.
3) Screen barley and wheat germplasm for resistance against barley/cereal yellow dwarf viruses.
4) Identify barley/cereal yellow dwarf virus-resistant genes and develop improved barley populations by introgressing identified resistance genes or QTLs into high yielding barley germplasm.
5) Evaluate wheat germplasm for resistance against wheat streak mosaic virus, Triticum mosaic virus, and barley yellow dwarf virus under field conditions.
Approach:
Barley yellow dwarf virus (BYDV) and cereal yellow dwarf virus (CYDV) are economically important viruses in barley in the USA. These viruses are transmitted by several cereal aphids in a persistent-circulative manner. Strain composition for BYDV and CYDV in wheat and barley is largely unknown in Nebraska. Field surveys will be conducted in Nebraska to collect symptomatic wheat and barley leaves from fields to determine the composition of BYDV and CYDV using RT-PCR with strain-specific primers. To define effective disease management strategies, it is important to understand the strain composition of BYDV (PAV, PAS, MAV, SGV) and CYDV (RPV, RPS) strains and co-infection with other major cereal viruses.
