Objective:
The ARS scientist and cooperator will develop developmentally relevant gene expression networks for non-infectious bud failure in almond. These networks will be used to identify the mechanism underlying bud failure and interrogated for markers and global patterns of gene expression diagnostic of disorder development.

Approach:
RNA sequencing will be done on vegetative buds of normal and diseased trees from the Stukey collection at the University of California Davis (UCD) Wolfskill orchard in Winters, California. Buds will be sampled over the spring to summer developmental trajectory from normal and affects shoots on diseased and normal trees. Expression and clustering will be done using an in-house pipeline, and identified clusters will be used for gene coexpression analysis using weighted gene co-expression network analysis (WGCNA). Differential network analysis will be conducted with DiffCorr.
