Objective:
1) Develop antiserum for the detection of Lettuce dieback associated virus (LDaV) that can be used for serological detection, 2) Clone LDaV and develop an agro-inoculation system for easier inoculation of plants during greenhouse evaluation of germplasm for LDaV resistance, and 3) determine if disease symptoms on lettuce are more severe when plants are infected by LDaV along with either of two tombusviruses previously known to be associated with the disease, or by LDaV alone.

Approach:
Antiserum will be developed against the virus nucleocapsid protein using an in vitro protein expression approach. The genome of LDaV will be cloned into agrobacterium vectors for use I agro-inoculation of lettuce for studies on host-plant resistance. Combinations of LDaV alone or LDaV with Moroccan pepper virus (MPV) and/or tomato bushy stunt virus (TBSV) will be inoculated to lettuce to determine the role of each virus and virus combinations in symptom development on lettuce.
