Interpretive Summary:

Technical Abstract: Temperature has been reported to significantly impact siRNA associated post transcriptional gene silencing (PTGS) of both viruses and transgenes in plants, suggesting that environmental conditions can have biologically important effects on RNAi associated pathogen defense and gene regulation. We have been using plant virus genes encoding suppressors of gene silencing and luciferase reporter genes to examine PTGS occurring during transient expression within Agrobacteria-infused leaf tissue. This assay appears to accurately reflect temperature effects on PTGS in plants and is being used to quantify the impact of other abiotic stresses (e.g. drought, nutrient, light) on gene silencing mechanisms in plants.
