Objective:
Objective 1: Test Grapevine Leafroll-associated virus 3 (GLRaV-3) resistance/tolerance in germplasm Rofar Vidor and C41-3,-5, -7, -24.
Objective 2: Investigate grape genes that are responsive to GLRaV-3 resistance/tolerance at the metabolomic level.
Approach:
Objective 1) The sample sizes of potential resistance varieties will be increased for larger-scale tests, and then challenged with virus inoculation. Virus will be inoculated by both mealybug transmission and mechanical transmission using the pure GLRaV-3 infectious clone, which can rule out the confusion that resistance may be caused by reduced mealybug preference.
Objective 2) Next, the downstream pathways of molecular grape-GLRaV-3 interaction will be investigated at the metabolomic level. Transcriptional dynamics that occur during plant-virus interactions must be translated into proteins and other metabolites to modulate viral pathogenesis. Several host factors influence viral pathogenesis at different stages of viral infection. As defense response signaling is mediated by different metabolites, we will determine the metabolomic outcomes of grape-GLRaV-3 interaction.
