Objective:
Investigate grapevine genes associated with grapevine leafroll-associated virus 3(GLRaV-3) resistance at the transcriptomic level.

Approach:
Objective 1) The sample sizes of potentially resistant varieties will be increased for larger-scale tests, and then challenged with virus inoculation. Virus will be inoculated by both mealybug transmission and mechanical transmission using the pure GLRaV-3 infectious clone, which can rule out the confusion that resistance may be caused by reduced mealybug preference. Objective 2) Next, the downstream pathways of molecular grapevine-GLRaV-3 interaction will be investigated at the transcriptomic level. Transcriptional dynamics that occur during plant-virus interactions must be translated into proteins and other metabolites to modulate viral pathogenesis. Several host factors influence viral pathogenesis at different stages of viral infection. As defense response signaling is mediated by different genes, we will determine the transcriptomic outcomes of grapevine-GLRaV-3 interaction.
