Objective I. Characterizing the Fungal Population. Sub-objective 1) Identify predominate fungal species associated with rotted fruits in the major production regions. Sub-objective 2) Investigate variations in fungicide sensitivity.
Objective 1-1: Sample cranberries from beds in Washington and Oregon at bloom and near harvest. Isolate fungi associated with various cranberry fruit rots from flowers and berries. Use polymerase chain reaction (PCR) assays on sampled tissues to detect specific species of fungi. Use multiplex PCR to identify cultured fungi. Objective 1-2: Isolates collected from cranberry beds in Washington and Oregon will be tested for resistance to representative fungicides from the Fungicide resistance action committee (FRAC) groups FRAC 3 chemistries and FRAC 11, which are the two major site-specific fungicide groups registered on cranberry. We will test members of the cranberry fruit rot complex that were isolated from cranberries collected from conventionally-managed cranberry beds and also organically-managed beds for resistance to fungicides.
