Objective:
Determine if the plant response to feeding is a function of potato variety, Lygus species (i.e., L. hesperus, L. elisus, L. lineolaris) geographic location of Lygus capture, and Lygus density on the potato plant. Compare the genetic responses of a Lygus “resistant” cultivar and a Lygus “susceptible” cultivar to feeding by field caught Lygus individuals using RNA sequencing. Characterize the salivary proteins in three Lygus species from multiple geographic locations (Oregon, North Carolina, and Wisconsin) using sachet feeding assays and mass spectrometry.
Approach:
Greenhouse studies to compare plant response as a function of variety and Lygus species. RNA sequencing to measure gene expression changes in potato caused by Lygus feeding. Proteomics to assess Lygus salivary proteins.
