Technical Abstract: Mitochondria are required for normal growth and development and play an important role in programmed cell death and aging. The mitochondrial DNA is maternally transmitted in the vast majority of eukaryotes. One exception is cucumber (Cucumis sativus), whose mitochondrial DNA is paternally transmitted. The differential transmission of the plant genomes in cucumber (maternal for chloroplast, paternal for mitochondrial and biparental for nuclear genes), the large mitochondrial genome and the existence of mosaic (MSC) mutants associated with major rearrangements in the mitochondrial DNA make the cucumber an excellent model for organellar genetics. A unique nuclear locus (Psm) control sorting of paternally transmitted mitochondria. We are using high resolution melt-curve analysis to perform fast and accurate genotyping of the large number of segregating progenies required for eventual cloning of Psm.
