Technical Abstract: Electroporation of barley pollen to transform barley with the Green Fluorescent Marker Protein (GFP) was studied. Transformed pollen containing the GFP gene, CaMV promoter and the Nos terminator fluoresced bright green when viewed under blue light (395 nm). Electroporation parameters affecting pollen viability, hence, transformation are field strength, capacitance and pulse length. DNA concentration and pollen stage had a significant impact on GFP activity.
