Title: Mapping of Fumonisin Biosynthetic Genes in Gibberella Fujikuroi

Technical Abstract: Gibberella fujikuroi mating population A (anamorph Fusarium moniliforme) causes stalk and ear rot of maize and produces a group of mycotoxins, fumonisins, that contaminate maize products. These toxins cause a number of animal diseases and are associated with human esophageal cancer. Fumonisins may also contribute to the virulence of G. fujikuroi on maize. We are examining the molecular genetics of fumonisin production via gene mapping and mutagenesis. We are mapping genes required for fumonisin production with RADS and amplified fragment length polymorphism. We are generating mutants of G. fujikuroi altered in fumonisin production by restriction enzyme mediated insertion, a procedure that greatly facilitates isolation of mutated genes. Since resistance to maize stalk and ear rot is currently unavailable, alternative control strategies should be examined. Understanding the molecular genetics of fumonisin biosynthesis may point to such strategies.
