Title: PROTEINS IN THE APHID SCHIZAPHIS GRAMINUM ASSOCIATED WITH VECTORING LUTEOVIRUSES

Technical Abstract: Luteoviruses are transmitted by aphids in a circulative, nonpropogative manner which is vector- and virus species-specific. Both aphid and virus structural proteins are involved in moving the virus through aphid gut and salivary tissues; the aphid proteins involved in virus transmission are unknown. 2D differential gel electrophoresis, co-immunoprecipitation, and tandem mass spectrometry were used to compare the proteomes of vector and nonvector genotypes of the aphid Schizaphis graminum. Over 60 differentially expressed proteins were identified between a vector and nonvector genotype. Examination of additional vector and nonvector genotypes identified that several proteins were exclusively associated only with vector genotypes. Co-immunoprecipitation of virus binding proteins from a total aphid protein mixture identified these same proteins in addition to several other proteins not identified in the differential proteome assay. These latter proteins may represent receptor or transport proteins found in minor amounts in the aphid. Mass spectrometry analysis determined the homology to some known proteins.
