Title: Characterization of the Maize Chlorotic Drawf Waikavirus Genome

Technical Abstract: Maize chlorotic dwarf waikavirus (MCDV) has 11.8 kb genome that encodes a single ca 400 kDa polyprotine. The polyprotine is post-translationally cleaved into several smaller functional proteins. Two Ohio isolates of MCDV incite different symptoms on susceptible maize. The "type" Ohio (MCDV-OH) isolate produces moderate symptoms, while the "severe" isolate (MCDV-S) produces much more severe symptoms on the same maize genotype. The sequenc of MCDV-OH is available (128269), as is that of a Tennessee isolate of MCDV (MCDV-TN). Although sequences for two MCDV isolates are available, there are three regions of the MCDV genome for which function has not been assigned. In order to characterize sequence differences among MCDV isolates and to determine the function of the three uncharacterized genomic regions, we synthesized and sequenced cDNA corresponding to regions of the MCDV-S genome. Preliminary data indicates that the MCDV-S sequence more than 90% identity with the MCDV-OH sequence. MCDV-S has significantly less similarity to the MCDV-TN and rice tungro spherical waikavirus (RTSV) sequences. His-tagged fusion proteins for cDNA corresponding to uncharacterized regions of the MCDV genome were expressed in E. coli and aare being used to raise antibodies in rabbits. These antisera will be to localize the MCDV-encoded proteins in the leafhopper vector, Graminella nigrifons andin maize.
