Title: Urocortin (Ucn) Decreased Feed Intake and Altered Serum Lh and Gh Concentrations in Pigs

Technical Abstract: Six ICV- and jugular-cannulated ovariectomized gilts 180 d of age were used in two replications to determine the effect of intracerebroventricular (ICV) injection of UCN on feed intake, GH, LH, ACTH, and cortisol (CS) secretion. Following a 20 h fast, feed was presented at 1 h after ICV injection (time= 0 h; during restraint) of saline (n=4; S) or 5 (n=3; U%) or 50 (n=5; U50) ug of UCN. Intake was assessed at 1.5, 2, 2.5, 3, 4, 6, 8, 10, 12 and 24 h. Blood was collected every 15 min for 2 h before injection, at 5, 10, 15, 20 and 30 min and every 15 min until 6 h for RIA of serum GH, LH, ACTH and CS. Sampling times were divided into periods of -2, 1, 2, 3, 4, 5 and 6 h relative to injection. Intake for U50 pigs decreased by 8 h and remained decreased for 24 h compared to S pigs (P<0.04), but was not altered in U5 pigs (treatment by period interaction, P<.05). A treatment by period interaction was detected for serum GH and LH Hconcentrations (P<.01). Serum GH was greater (P<.001) at 1 h for U5 and 1 and 2 h for U50 compared to -2 h, but was not influenced by S. Compared to -2 h, S gilts had lower serum LH concentrations (P<.002) at 1 h while U5 gilts had lower (P<.02) serum LH concentrations at 2, 5 and 6 h when compared to -2 h for U5 and U50. Serum ACTH and CS concentrations were not influenced by treatment. These data provide evidence that UCN modulates GH and LH secretion and suppresses feed intake in the pig which may be independent of ACTH and CS release.
