LOCATING CUCUMBER AND CARROT DNA SEQUENCE IN CHROMOSOMES

Evaluate carrot and cucumber genetic marker relationships to chromosomes.
1b.Approach (from AD-416)
1) Compare linkage map location to marker sequence for 15 genes each in carrot and cucumber.
2) Compare carrot and cucumber marker sequence to GenBank and other data bases.
3) Evaluate sequences for carrot in BAC library.
Over 100 deoxyribonucleic acid (DNA) sequences were located on chromosomes and compared to DNA sequence maps generated from whole genome sequencing. Excellent correlation between genetic and physical location was observed. Additional sequence for flanking sequence was gathered from carrot bacterial artificial chromosome (BAC) library, and sequence polymorphism useful for chromosome mapping was discovered. This data is of interest to plant geneticists and will be used to assemble larger sequence datasets. Project monitored by regular meetings and e-mail discussions of progress.
