Interpretive Summary: Erigeron breviscapus is a whole plant used in traditional Chinese medicine for treating cardiocerebral diseases. A standardized method was used to identify and quantify 33 phenolic acids and 16 flavonoids. A similar analysis of breviscapine, an impure compound-enriched residue of Erigeron breviscapus used as a therapeutic drug, revealed 18 flavonoids. Quantification was accomplished using 2 reference standards and molar relative response factors developed at ARS. The results of this study offer the possibility of understanding the bioactivities of this herb more clearly and of using it more efficiently in the future.

Technical Abstract: A standardized LC-PDA-ESI/MS profiling method was used to identify the phenolic components in Erigeron breviscapus and a general phenol quantification method was used to quantify each component. Thirty-three hydroxycinnamates and 16 flavonoids were found in Erigeron breviscapus, a whole plant used in traditional Chinese medicine for treating cardiocerebral diseases, and 18 flavonoids were found in the impure compound-enriched residue of breviscapine, a derived therapeutic drug. The hydroxycinnamates were quantified using the peak area at 326 nm, dried chlorogenic acid as a calibration standard, and the mole relative response factors (MRRF) for each compound. Similarly, 8 apigenins, 4 luteolins, 3 flavonol 3-glycosides, and the 7-O-glucuronides of quercetin and isorhamnetin were quantified using apigenin (at 336 nm) and rutin (at 354 nm) as calibration standards and MRRF value for each compound. The results of this study offer the possibility of understanding the bioactivities of this herb more clearly and of using it more efficiently in the future.
