Interpretive Summary:

Technical Abstract: Bovine viral diarrhea viruses (BVDV) have recently been segregated into two genotypes: BVDV 1 and BVDV 2. In this study, we have begun to define the antigenic similarities and differences of BVDV 1 and BVDV 2 with two neutralizing anti-E2 monoclonal antibodies (mAbs) produced to an isolate of BVDV 1. The mAb 157 was found to be BVDV 1-specific, whereas mAb 348 bound dto and neutralized BVDV 2. Both mAbs bound to a C-truncated E2 protein (amino acids [aa] 1 to 192). By sequencing of neutralization escape mutants, the epitope of mAb 157 was found to be affected by changes at aa positions, 9, 32, and 72. By alignment of sequences, BVDV 2 are missing the equivalent of aa 31 and 32 of BVDV 1. The epitope of the type-common mAb 348 was affected by changes at aa positions 71, 72, 74, and 77.
