Experiments are being conducted to improve tissue culture systems for malting barley. Changes in the chemical composition of the tissue culture media have increased green plant regeneration three to ten fold in malting barley. Sucrose can be substituted for maltose as early as four weeks after culture initiation, providing major reductions in the costs of tissue culture. Autoclaving media in separate parts rather than in one flask provided a two to three fold increase in regeneration. The effects of donor plant growth environment also are being investigated with the goal of identifying the optimal greenhouse planting time to maximize regeneration from Morex. Results from these experiments will identify optimal tissue culture systems for transformation of malting barley cultivars. |
