Interpretive Summary:

Technical Abstract: A new siderophore, AMG-NS-16a, was isolated from the fermentation of culture AMG-NS-16, recovered from an inclusion in a 65 million- year-old amber piece from the Raritan formation in New Jersey. It is possible, however, that the isolate might not be as old as the amber sample. The isolate AMG-NS-16 was tentatively identified as a new strain of Streptomyces griseus based on its morphological and physiological properties. The fermentation of AMG-NS-16 was conducted in a medium containing soluble starch (20 g/l), glucose (10 g/l), peptone (5 g/l), yeast extract (5 g/l), and calcium carbonate (5 g/l), using a 18-h seed prepared in the medium ATCC#172. The AMG-NS-16a contained in the fermentation filtrate was recovered by cation-exchange chromatography and further purified by size-exclusion chromatography. Spectroscopic analysis, including HRFAB-MS and 2D-NMR identified its chemical structure to be related to the aerobactins.
