Interpretive Summary:

Technical Abstract: The pituitary plays significant roles in the regulation of a wide variety of physiological processes in the body. In the current study, we obtained expressed sequence tag data to characterize the transcriptome of the rainbow trout pituitary. Clones (n=1920) from a normalized cDNA library constructed from pooled male and female pituitaries were sequenced to identify as many unique transcripts as possible. From these 3840 sequences, a total of 524 contigs were assembled and 1256 singletons identified. Comparison to the Rainbow Trout Gene Index revealed that 77.3% of the contigs and 78.9% of the singletons are novel sequences and represent a significant addition to the existing complement of expressed sequence tag data for rainbow trout. Transcripts were functionally annotated using BLAST comparison to the non-redundant database at NCBI and gene ontology term assignment using the GOfigure gene ontology annotator. Digital Differential Display found the pituitary hormones and extra-cellular matrix proteins to be the most abundant transcripts. Through in silico comparative mapping homologs were identified for 354 of the unigene sequences. These data provide the first functional and mapping information on many of the transcripts present in the rainbow trout pituitary.
