Technical Abstract: Pistia stratiotes L. produces calcium (Ca) oxalate crystals in specialized cells called crystal idioblasts. Druse crystal idioblasts are produced in the adaxial mesophyll and raphide idioblasts are produced in the abaxial aerenchyma of this aquatic plant. Leaves formed on plants grown on 0 Ca medium have fewer and smaller druse crystals than leaves formed when plants are grown on media supplemented with 5 or 15 mM Ca. Druse crystal formation was reduced when plants were treated with Ca channel blockers such as isradipine, nifedipine or fluspirilene. In protoplasts, 43Ca uptake was more than two orders of magnitude greater for crystal idioblast protoplasts compared to mesophyll protoplasts and an antibody to a mammalian Ca channel alpha-1 subunit heavily labeled developing crystal idioblasts in tissue sections. The results demonstrate that excess Ca can be stored as Ca oxalate and that dihydropyridine-type Ca channels play a key role in the transport and accumulation of Ca required for crystal formation.
